Experimental principle:
Filter a certain amount of sample with a filter membrane to trap the algae, grind and break the algae cells, extract the chlorophyll with acetone solution, measure the absorbance of the extract at 750nm, 664nm, 647nm and 630nm wavelengths after centrifugal separation, and calculate the concentration of chlorophyll a in the water according to the formula.
Application of JXFSTPRP-YLS standard method for extracting chlorophyll:
1. Filtration: Add magnesium carbonate suspension before suction filtration to prevent pigment decomposition. The negative pressure cannot exceed 50kPa.
2. Chlorophyll extraction: place the filter membrane in the JXFSTPRP-YLS extractor, add 10ml of 90% acetone solution, and grind for 1min at 1500-2000rpm. Break the phytoplankton algae cells on the filter membrane. After grinding, it is soaked and extracted in the dark at 4℃, and the soaking time is 2h to 24h. Final centrifugation and clarification: Centrifuge at 1000g for 20 minutes or filter, and transfer the supernatant to a stopcock glass vial.
3. Determination method: spectrophotometry, three-color equation to determine the absorbance of 664nm, 647nm, 630nm, 750nm to calculate the chlorophyll a, b, c chlorophyll content. Determination of the corrected content of pheophytin a by monochromatic equation
JXFSTPRP-YLS can process 18×15 mL centrifuge tubes at the same time for rapid cell disruption, cell lysis, and tissue homogenization.
